    摘要:目的通过检测喉鳞状细胞癌(laryngeal squamous cell carcinoma)患者肿瘤组织中吲哚胺2,3双加氧酶(indoleamine 2,3dioxygenase,IDO)和桥接整合因子1(bridging integrator 1,BIN1)的表达,探讨其在喉癌发生发展中的作用。方法应用免疫组织化学染色法检测50例喉鳞状细胞癌肿瘤组织和23例正常喉黏膜组织中IDO和BIN1的表达,并分析两者表达的相关性及其与患者临床特征的关系。结果IDO在50例喉癌中阳性表达率为60%,而在癌旁正常喉黏膜组织中IDO表达为17.39%,与癌旁正常喉黏膜组织比较,喉癌IDO阳性表达率明显增高,差异具有统计学意义(P<0.01);BIN1在50例喉癌中阳性表达率为50%,而在癌旁正常喉黏膜组织中BIN1蛋白阳性表达率为95.65%,与癌旁正常喉黏膜组织比较,BIN1阳性表达率明显降低,差异具有统计学意义(P<0.01);而且IDO在原发灶和转移灶中的表达显著增高,BIN1在原发灶和转移灶中的表达降低,差异具有统计学意义(P<0.05)。在喉癌肿瘤组织中,IDO表达与有无淋巴结转移、临床分期相关;BIN1表达与肿瘤分化程度、有无淋巴结转移、临床分期相关。结论喉癌患者肿瘤组织和颈部转移淋巴结中IDO表达水平明显提高,而BIN1表达水平显著降低,并且与患者临床特征紧密相关,这两者可能是影响喉癌发生发展的重要因素。

    Abstract:ObjectiveTo study the expressions of indoleamine 2,3dioxygenase (IDO) and bridging integrator 1(BIN1) in laryngeal squamous cell carcinoma, and to analyze their clinicopathological significances.MethodsThe expressions of IDO and BIN1 in 50 specimens of laryngeal carcinoma and 23 of normal laryngeal mucous tissues were detected by immunohistochemical staining, and correlation between the two expressions and their relations with clinical characteristics of the patients were analyzed.ResultsThe positive expression rates of IDO were 60% and 17.39% in specimens of laryngeal carcinoma and normal laryngeal mucous tissue respectively, and the difference was statistically significant (P<0.01). The positive expression rates of BIN1 in specimens of laryngeal carcinoma and normal laryngeal mucous tissue were 50% and 95.65%, and the difference was also statistically significant (P<0.01). Comparing primary tumors with metastatic ones,the expression level of IDO was upregulated, while that of BIN1 was downregulated with statistically significant differences (both P<0.05). The expression of IDO protein was associated with lymph node metastasis and clinical stage of the tumor,while the expression of BIN1 protein was associated with degree of differentiation, lymph node metastasis and clinical stage of the tumor.ConclusionsIn laryngeal squamous cell carcinoma specimens and metastatic cervical lymph nodes,the expression level of IDO is upregulated significantly and that of BIN1 is downregulated,which is closely associated with clinical features of the tumor. IDO and BIN1 might be important factors affecting occurrence and progression of laryngeal squamous cell carcinoma.
